چکیده

The potential of many drugs and chemicals to cause prenatal harm is well-established. The types of effects seen vary and include spontaneous abortions, stillbirths, congenital malformations present at birth, and conditions detected only months to years after birth. Most studies designed to determine whether agents cause malformations have examined the effects of exposure of the embryo or fetus at various times during gestation. Thus the effects studied are maternallymediated. Despite these drug studies and those on other causes of birth defects, such as chromosomal abnormalities, the cause of approximately 60% of congenital malformations is unknown. Fewer studies have examined the possibility that exposure of the male to a drug or chemical could lead to abnormalities in his offspring, i.e., be maleor paternally-mediated. Over the last 10–20 years, there has been more interest in addressing this question as increasing numbers of patients and workers are exposed to agents that alter fertility. A number of animal studies as well as more recent human epidemiological studies have demonstrated that exposure of males to various agents can result in abnormal reproductive, pregnancy, and/or progeny outcomes (Olshan and Mattison, ’94).

Cross-species sperm-FISH assays for chemical testing and assessing paternal risk for chromosomally abnormal pregnancies.

The father, like the mother, can transmit genetic defects to his offspring that are detrimental for normal development and a healthy life. Epidemiological studies have identified associations between several paternal exposures and abnormal reproductive outcomes, but these types of studies are inherently complex and expensive, and the risk factors for the paternal contribution to abnormal reprod...

Relative susceptibilities of male germ cells to genetic defects induced by cancer chemotherapies.

Some chemotherapy regimens include agents that are mutagenic or clastogenic in model systems. This raises concerns that cancer survivors who were treated before or during their reproductive years may be at increased risks for abnormal reproductive outcomes. However, the available data from offspring of cancer survivors are limited, representing diverse cancers, therapies, time to pregnancies, a...

Asbestos-related Lung Diseases: A Brief Update

Health risks from asbestos exposures have been evaluated, considering past professional histories when exposures at workplaces were higher than today. A linear no-threshold (LNT) model has been applied, although its relevance is unproven. Fibers are often found in the lungs and pleura of deceased people. Fiber findings do not prove that a disease is caused by asbestos. It is reasonable to assum...

Evidence for a role of paternal exposures in developmental toxicity.

Experimental evidence from radiation exposure, antimitotic drugs or chemicals such as pesticides or metals does suggest the possibility of transmission of paternally mediated developmental effects across generations. The mechanistic framework is growing with suggestion of transmission of epigenetic modifications as a mechanism alternative to germ-line mutagenesis. There is also ample experiment...

Paternal Risk Factors for Oral Clefts in Northern Africans, Southeast Asians, and Central Americans

While several studies have investigated maternal exposures as risk factors for oral clefts, few have examined paternal factors. We conducted an international multi-centered case-control study to better understand paternal risk exposures for oral clefts (cases = 392 and controls = 234). Participants were recruited from local hospitals and oral cleft repair surgical missions in Vietnam, the Phili...
